In Story mode, Chamber 01 (also known as Virgil-01) is the second map of the third chapter and the first to be recognized as a test chamber.
This chamber also has a Ratman's Den in the game (which is related to the achievement Under the Stairs). The map is divided in two tests.
The first one makes use of Goo and introduces the player to the Excursion Funnels. In Advanced mode, it also introduces Laser Fields.
The second one, in Advanced mode, makes use of Propulsion Gel.

Trivia
- There is a secret room with art similar to those by Doug Rattmann in this chamber. There are two murals depicting AEGIS and Mel’s antique Portal Gun. Virgil also says that he has heard rumors about a creepy guy living in the service areas.
